Abstract
Objective To investigate the effects of Alprostadil injection on insulin resistance and TLRs (toll like receptor) signaling pathway in peripheral blood mononuclear cells of patients with diabetes mellitus.Methods 80 patients with diabetes were divided into observational group and control group,based on whether accept the Alprostadil treatment,40 patients in each group.Patients in the two groups were all given conventional diabetes treatment.The effect of the index of glucose metabolism,renal function and inflammatory factors on TLRs signaling pathway in peripheral blood mononuclear cells were compared between the two group.Results After treatment,TLR2 (toll like receptor 2),TLR4 (toll like receptor 4) and NF-κB (nuclear factor κB) levels in patients of the observational group were 19.7 ± 6.3,4.7 ± 1.7 and 8.6 ± 2.4,which were significantly lower than that in the control group(TLR2:t =-8.09,P =0.00;TLR4:t =-8.52,P =0.00;NF-κB:t =-8.09,P =0.00).The levels of GLU (mmol/L),FIN (μg/L),HOMA-IR,HbA1c (%) were 5.36 ± 0.67,3.94 ± 0.57,1.86 ± 0.54,3.67 ± 0.65,which were higher than that in the control group(t =-6.51,P =0.00;t =8.53,P =0.00;HOMA-IR:t =-6.96,P =0.00;HbA1c:t =11.87,P =0.00).The levels of urea nitrogen,urine protein,β2 MG were 5.61 ± 1.89,0.59 ± 0.16,2.92 ± 0.61,which were significantly higher than that in the control group(urea nitrogen:t =-2.07,P =0.04;urine protein:t =-8.65,P =0.00;β2-MG:t =-13.89,P =0.00).TNF-α,IL-6,IL-8 levels were 16.8 ± 2.3,4.7 ± 0.6,5.9 ± 0.5,which were significantly lower than that in the control group(TNF-α:t =-18.67,P =0.00;IL-6:t =-5.40,P=0.00;IL-8:t =-8.09,P=0.00).Conclusion Alprostadil injection could inhibit the activation of TLRs signaling pathway and improve insulin resistance in diabetic patients.关键词
