P38MAPK Expression in Motor Neuron of Anterior Horn of Lumber Spinal Cord under the Treatment of Electro-acupuncture on Jiaji Points in ALS-SOD1G93A Transgenic Mice

Abstract
Objective:To observe the expression of P38,P-P38 in motor neuron of anterior horn of lumber spinal cord by electro-acupuncture on Jiaji points in ALS-SOD1G93A transgenic mice,to prove acupuncture protective effect on neuron and to provide experimental basis for the acupuncture treatment for ALS.Methods:18 ALS-SOD1G93A transgenic mice were randomly divided into the electro-acupuncture group,the acupuncture group,the model group,with six mice in each group;six wild-type mice were taken as negative control.The mice were intervened starting from 60 days after birth.The electro-acupuncture group was intervened with rarefaction wave on Jiaji points of L1-2,and L5~6;the acupuncture group was intervened with hand acupuncture on Jiaji points of L1~2,and L5~6,the frequency for the needle twirling was once every ten minutes;the model group and the negative control group were intervened with grasping and seizing at the same time for the same time length.The treatment course was four weeks.120 days after birth,the mice were given immunohistochemical test,and the mean density and IOD were analyzed by image analyzer.Results:The expressions of P-P38 and P38,and the ratio of P-P38/P38 in the model group were significantly increased compared to the negative control group(P <0.01).The expressions was decreased in the electro-acupuncture group compared to those of the model group and the acupuncture group(P <0.01 or P <0.05),while there were no significant differences between the acupuncture group and the model group (P > 0.05).It proved that electro-acupuncture could reduce the expressions of P-P38 and P38,as well as the ratio of P-P38/P38.Conclusion:Electro-acupuncture on Jiaji points can down-regulate the expression of P-P38 in motor neuron of anterior horn of lumber spinal cord in the middle age of ALS-SOD1G93A transgenic mice,and inhibit the activation of P38MAPK to protect the neuron.关键词
