摘要

Abstract

Objective To investigate effect of Pingchuan granule on the expression of epidermal growth factor protein in asthmatic rats after airway remodeling by measuring the expression of epidermal growth factor protein after airway remodeling in asthmatic rats. Methods 90 Wistar rats were divided into 6 groups, 15 in each group, group A and group B were given normal saline by intragastric administration. Group C was inhaled with budesonide (0.5 mg of budesonide suspension, inhaled in compressed atomizer, twice daily), D, E and F were given low dose (4.01 g /kg/d), medium (8.01 g/kg/d) and high (16.02 g/kg/d). Each group was sacrificed 24 hours after the last excitation, and the lung tissue of the middle and lower part of the left lung was cut into 1mm × 1mm × 1mm size. The tissue was placed in an EP tube with 2.5% glutaraldehyde and fixed for electron microscopy. Take part of the right lung tissue, PBS rinse dry water, liquid nitrogen frozen, placed in -80 ℃ refrigerator for RT-PCR detection. Results The expression of EGF mRNA protein in (the low、medium、high) dose Pingchuan granule group and Budesonide group and model group was higher than that in the blank group, and the difference was statistically significant (P < 0.05). The expression of EGF mRNA protein in (the low, medium, high) dose Pingchuan granule group and budesonide group was significantly lower than that in model group (P < 0.05). The expression of EGF mRNA protein in (the medium, high) dose Pingchuan granule group was significantly lower than that in budesonide group but with no statis tical difference (P > 0.05). The expression of EGF mRNA protein in low dose Pingchuan granules group was lower than that in budesonide group, but the difference was not statistically significant. Conclusion Pingchuan granule can improve and control the airway remodeling process of airway hyperresponsiveness and airway obstruction status, and can inhibit the expression of EGF mRNA, thereby protecting the lung tissue cells, reduce asthma attacks.
