Abstract
Objective:To investigate the effect of Radix Hedysari polysaccharide (Hedysarum Polybotys saccharide,HPS)on streptozotocin-induced diabetic rat model of retinal vascular endothelial growth factor (vascular endothelial,growth factor,VEGF) and pigment epithelium derived factor (Pigment epithlium-derived,facto,PEDF) and serum C-reaction protein (Creactive protein,CRP) level and the effect of retinal histopathological changes,and research the mechanism of Radix Hedysari.Methods:150healthy rats were randomly divided into experimental group and controlled group,25rats in the controlled group received intraperitoneal injection of citrate buffer,125rats in the experimental group received intraperitoneal injection of streptozotocin.The experimental group animal model was successfully divided into HPS low dose group,HPS medium dose group,HPS high dose group and blank group,Dobe group,the HPS groups rats were given 50mg/kg,100mg/kg,150mg/kg concentration of HPS gavage,the normal saline group and the blank controlled group were given intragastric administration Dobe,Dobe,90mg/kg group was given orally,1 times/day,orally for 8weeks.The last time after gavage,the serum of the rats to detect CRP content,and the removal of fixed rat eye on formaldehyde solution,and then stripping the retina pathological observation and detection of retinal tissue VEGF and PEDF levels.Results:The serum CRP test results:compared with the controlled group,serum CRP content of rats in the blank group,the difference was statistically significant (P<0.05);compared with the controlled group,the HPS groups and calciumdobesilate group serum CRP levels were decreased,the difference was statistically significant (P<0.05);and more than the Adams group,high,medium and low the concentration of HPS group rats serum CRP content decreased significantly (P < 0.05);Pathological observation:compared with the controlled group,HPS rats retinal damage decreased;Compared with the most obvious HPS calciumdobesilate group,high dose group of rat retinal thickening,each layer of cells were arranged in order.The results of PEDF and VEGF:compared with the controlled group,increased the level of VEGF in retinal tissue in the controlled group,PEDF content decreased,the difference was statistically significant (P<0.05);Compared with the controlled group,the HPS groups and the VEGF group each dose of calciumdobesilate levels decreased,PEDF content increased,the difference was statistically significant (P<0.05).Conclusion:Radix Hedysari polysaccharide can reduce serum CRP content in rats is to reduce the level of VEGF in retinal tissue,the content of PEDF increased,curb the growth of new blood vessels and retinal to protect retinal function.关键词
