芍药苷预处理对微血管内皮细胞缺氧复氧模型的干预作用
Intervention effects of pretreatment with paeoniflorin on microvascular endothelial cell hypoxia-reoxygenation model

Abstract
Objective To explore the intervention effects of pretreatment with paeoniflorin on microvascular endothelial cell hypoxia-reoxygenation model.Methods Microvascular endothelial cell hypoxia-reoxygenation models of rats were prepared and randomly divided into five groups,i.e.a control group (blank control),a hypoxia-reoxygenation group (treated with ischemiareoxygenation),and three paeoniflorin pretreatment groups (treated with ischemia-reoxygenation +low,medium and high doses of paeoniflorin for pretreatment).Flow cytometry (FCM) was used to detect the cell apoptosis in each group,and ELISA method was applied to detecting the expression of TNFα and IL-1β.Results Compared with the hypoxia-reoxygenation group,the levels of TNFα and IL-1β in the three dosage groups of paeoniflorin pretreatment were significantly lower (P < 0.01),but there was no significant statistical difference among the three groups (P > 0.05);the detection results of FCM snowed that the apoptosis index in the paeoniflorin pretreatment groups was significantly lower than that in the hypoxia-reoxygenation group (P < 0.05),but there was no significant statistical difference among the three groups,either (P > 0.05).Conclusion Pretreatment with paeoniflorin can reduce the apoptosis of microvascular endothelial cell in rats induced by ischemia-reoxygenation and protect microvascular endothelial cell.This may be correlated to the reduction of the expression of TNFα and IL-1β.关键词
