Abstract
Objective To establish a method for detection of Diego blood group gene by Touchdown PCR,and to analyze the distribution of Diego blood group polymorphism of blood donors in Xi'an area,and to establish the rare blood group pool of Diego.Methods Diego gene was detected by Touchdown PCR,and anti-Dia serum and commercial genotyping kits were used to verify the results,then sequenced.Genomic DNA was extracted from 1 068 blood donors in Xi'an area,and Diego blood group genotyping was performed.Results The Touchdown PCR method for the detection of Diego blood group gene was established.1 Di(a+b-),52 Di (a+b+),1 015 Di (a-b+) and no Di (a-b-) were detected in all of the samples.In blood donors,the expression frequency of Dia antigen was 0.04963,and Dib antigen was 0.99906.Conclusion In our study,we successfully established a method to detect the Diego blood group gene by touchdown PCR,and screened and analyzed of Diego blood group of blood donors in Xi'an area,and obtained the data of its polymorphism distribution.关键词
