牛ACOX1基因过表达载体构建及对脂肪沉积的初步研究
Construction of Over-expression Vector of Bovine ACOX1 Gene and Preliminary Study on Fat Deposition

Abstract
In order to elucidate the role of bovine acyl-CoA oxidase 1 (ACOX1) in intramuscular fat deposition,and provide the theoretical basis for the cultivation of high quality beef cattle breeds,the CDS sequence of bovine ACOX1 gene was amplified and the over-expression vector of ACOX1 was constructed firstly in this study,and then the over-expression plasmid was transfeted into 3T3-L 1 cells.Lastly,the deposition of lipid droplets was detected by oil red O staining and the expression levels of PPARy and C/EBPα were detected by fluorescence quantitative PCR and Western blotting.BLAST analysis and digestion identification showed that the over-expression vector of bovine ACOX1 gene was successfully constructed;the results of oil red O staining showed that over-expression ofACOX1 gene inhibited lipid droplet deposition in 3T3-L1 cells;the results of fluorescence quantitative PCR and Western blotting showed that over-expression of ACOX1 significantly inhibited C/EBPα expression but had no significant effect on the expression of PPARy.These data suggest that ACOX1 may inhibit the deposition of intramuscular fat.关键词
