黄芪配方颗粒对衰老大鼠认知功能的影响及机制初探
Explore the Effect and Mechanism of Astragalus Particles in Aging Rats with Cognitive Impairment

Abstract
Objective:To study the effect and mechanism of Astragalus particles on improving memory and cognitive effects.Methods:Rats were randomly divided into control group,donepezil hydmchloride group (0.5 mg/kg),high-dose Astragalus particles group (HQKL-H,0.450 g/kg),moderate-dose Astragalus particles group(HQKL-M,0.275 g/kg)and low-dose Astragalus particles group (HQKL-L,0.138 g/kg).Rats were given correspond drugs for continuous 12 w.After administrated for 10 w,the space memory capacity was detected by the test of Morris water maze.Rats were executed after administrated for 12 w,the autonomic activities were measured.The levels of SOD,MDA and CAT in serum and brain were detected by biochemical process and ELISA assay.The morphological changes in hippocampus tissue were detected by HE staining,and the positive expression of Nrf2 was measured by immunohistochemical method,the protein expression level of Nrf2 and HO-1 were measured by Western blot.Resuhs:HQKL-H could significantly increase the autonomic activities,the level of SOD and CAT in serum and brain were significantly increased,the protein expression of HO-1 and Nrf2 were significantly increased(P <0.05 or P <0.01),while the escape latency,total distance and time in the fourth quadrant,and the level of MDA were significantly decreased.Compared with aging model group,HQKL-H significantly increased the protein positive expression of Nrf2 in brain,and improving the nerve cell in hippocampus tissue.Conclusion:Astragalus particles can improve the learning capacity,the mechanism may be relevant to suppress the lesion in hippocampus and the anti-oxidative pathway of Nrf2/ARE.关键词
