摘要

Abstract

The expression of dairy goat ovary miRNAs was analyzed by Solexa/Illumina high-throughput sequencing technology,which producing same-sex lambs,thus providing a basis for exploring the role of miRNAs in the regulation of fertility.The total RNA was extracted from the ovaries of the dairy goat mothers with same-sex-lambs.The eDNA library was established and cloned,sequenced,and conducted bioinformatics analysis.The results show there were 744,782 miRNAs detected in the goat ovary which produced same-male-lamb (ASGO1),while 1602751 miRNAs were detected in the goat ovary which produced same-famale-lamb(ASGO2).There were 630 species of miRNAs expressed differentially between ASGO1 and ASGO2.143 species expressed only in the ovary which produced same-famale-lamb (ASGO2).70 species expressed only in the ovary which produced same-male-lamb (ASGO1).There are 31 new miRNAs.Two miRNAs (ssc-miR-451_R-1 and bta-miR-486_R-1) in ASGO2 were significantly different from ASGO1.Bta-miR-486_R-1 was the largest difference,reaching a high expression level of 800,000 copies,Ssc-miR-451_R-1 and bta-miR-486_R-1 are likely to be important miRNAs that affect selective fertilization of dairy goats.
