Abstract

In this study,a MsHSC70-2 gene was determined by polymerase chain reaction (PCR),and its mRNA sequence is 2025bp,which contains a complete open reading box of 1950bp,encoding 649 amino acids,molecular weight of the protein is about 70.97 Kda,its isoelectric point is about 4.74.The mRNA content of MsHSC70-2 from different organs of the variety in different seasons were detected by qRT-PCR,showing that its content in the same organs had significant difference in different seasons(P<0.05),and the content of MsHSC70-2 mRNA among organs was different during summer(P<0.01).The expression trend of the gene in root,stem,leaf and bud increased first and then decreased from April to September,demonstrating that the expression of MsHSC70-2 is regulated by day length and temperature,and its expression variation affects the growth and development of various organs,its high expression promotes the growth and development of alfalfa,and plant growth was inhibited at low expression level,showing that decrease of its expression may participate in fall dormancy in alfalfa.The study provides significant basis for further study on the function of alfalfa HSC70-2.
