摘要

Abstract

In this paper, the expression stability of the three common house-keeping genes in the hypothalamus and ovary of chicken at different stages of development was analyzed, this provided the scientific basis for the selection of quantitative detection of related functional genes on gonadal axis of chickens. Jining Bairi chicken at different sexual development stages as experimental material, used the real-time fluorescence quantitative PCR detecting common housekeeping gene actin gene beta-(beta-actin), 3-glyceraldehyde-3-phosphate dehydrogenase gene (glyceraldehyde-3-phosphate dehydrogenase GAPDH) and 18S ribosomal RNA gene (18S rRNA) expression in hypothalamus and ovarian tissue. The softwares of geNorm, NormFinder and BestKeeper were used to compare the stability of these genes expression. The results showed at the different stages of sexual development of Jining Bairi chicken, GAPDH gene was the most stable expression in the hypothalamus, and the most stable expression in the ovaries was the beta actin gene. The results provide a reference for the accurate analysis of the relative expression of genes on the HPG of chicken by real-time fluorescence quantitative PCR.
