Abstract

Objective:To investigate the expression of Thrombospondin-2(THBS2)and its effect on proliferation,migration and invasion during the development of cervical cancer. Methods:The expression of THBS2 in tissue or cell lines was detected by immunohistochemis-try and Western blot. Expression plasmid was used to increase the level of THBS2 in cervical cancer cell lines-SiHa and ME180,and siRNA was used to decrease the expression of THBS2 in H8 and ECT-1 cells. The results were verified using Western blot. CCK8 assay,wound healing and Transwell invasion assay were performed to estimate the effect of THBS2 on proliferation, migration and invasion in cervical cancer cell lines. Results:THBS2 was down-regulated during the development of cervical cancer (P<0. 001). And THBS2 in cervical cancer cell lines SiHa and ME180 was significantly lower than those of H8 and Ect-1(P<0. 05). The proliferation (P<0. 05),migration (P<0. 005)and invasion (P<0. 001)effects were inhibited in SiHa and ME180 cell lines when THBS2 was overexpressed. The proliferation (P<0. 05),migration (P<0. 005)and invasion (P<0. 001)effects were promoted in H8 and ECT-1 cells when THBS2 was suppressed. Conclusion:THBS2 plays a role as 'cancer suppressive factor' during the de-velopment of cervical cancer and thus may be a potential therapeutic target for treating cervical cancer.
