猪圆环病毒3型PCR诊断方法的建立
Establishment of PCR diagnostic method for porcine circovirus serotype 3

Abstract
In 2016 it was first reported that a new serotype of porcine circovirus was appeared as porcine circovirus type 3 (PCV3)in the United States.In order to rapiddly diagnose PCV3,the PCR diagnostic methodon PCV3 was established.According to the genome sequence of PCV3 from NCBI,a pair of specific primers was designed and the reaction conditions of PCR were optimized,and the specificity and sensitivity of PCR were carried out.Finally 22 clinical cases were detected by this PCR diagnostic method.The results showed that the total volume of PCR was 25 μL.The annealed temperature was at 56 ℃,the amount of template DNA was 3 μL (1 ng/mL)and optimized cycles were 35 cycles.In the optimum condition,a specific fragment of about 381 bp was amplified.The results of sequencing showed that the production of PCR was correct.The results of specific test showed that the PCR reaction only amplified the PCV3 positive samples.The results of sensitivity test showed that the PCR reaction was able to amplify the lowest amount of PCV3 positive with templates of 1 × 10-4 ng/mL.The positive rate of PCV3 from 22 clinical cases was 13.6%.Therefore,the PCR diagnosis method on PCV3 is specific,sensitive,and can be used for the rapid detection of PCV3.关键词
