摘要
Abstract
The technique of separation and purification of 2′-deoxyadenosine by macroporous adsorption resin was systematically investigated.The adsorption capacity of seven different kinds of resins were studied based on the static adsorption. The results showed that D101 resin was the optimal sorbent and the procedure of desorption were sequential filtration with 0.1 mol/L Na2 CO3 solution and 30% ethanol at pH 8.0.The optimum operating parameters were determined by dynamic adsorption as follows :sample flow rate of 2.0 mL/min ,elution flow rate of 1.5 mL/min ,eluting with 4.5 times column volume of 0.1 mol/L Na2 CO3 ,3 times column volume of 30% ethanol orderly. Under the condition ,the content of 2-deoxyadenosine is as high as 78.7% ,and the yield could reach t o 93.9%.关键词
