Abstract
Objective To investigate the effect of RNA interference(RNAi)of B-cell specific moloney murine leukemia virus insertion site1(Bmi-1)gene in human squamous cell carcinoma cell line A431 on its proliferation.Methods ]RNAi of Bmi-1 gene was transfected to squamous cell carcinoma cell line A431, and the Bmi-1-RNAi silencing vectors of A431 cell lines were screened by reverse transcription polymerase chain reaction(RT-PCR).The expression of Bmi-1 protein was determined by Western blotting, and the inhibition of cell proliferation was detected by using methyl thiazolyl tetrazolium(MTT).Cell cycle was flow cytometrically determined.Results Two days af-ter transfection with Bmi-1 shRNA, the A431 cells became small, the gap became larger, and the growth rate slowed down.The expression of Bmi-mRNA was significantly lower in Bmi-1 shRNA transfection group than in control group(P<0.05).The expression of Bmi-1 protein in the Bmi-1 shRNA transfection group was significantly lower than that in the blank control group.The percentage of S phase cells was higher and the percentage of G2/M phase cells was lower in Bmi-1 shRNA transfection group than in blank control group(P<0.05).Conclusion RNAi can down-regulate the expression of Bmi-1 mRNA and protein, inhibit the differentiation and proliferation of A431 cells and block their growth cycle.关键词
