黄连属植物DNA条形码研究
DNA Barcoding Evaluation in the Genus Coptis Salisb.

Abstract
The present study made a global samling of Coptis and selected four frequently used DNA bar-codes ( trnH-psbA and ycf1 from cpDNA,ITS and ETS from nrDNA) ,to evaluate their species discrimination a-bility in the genus.Based on the comprehensive analyses of the sequence characteristics,intra/inter-specific ge-netic distance,DNA barcoding gap and neighbor-joining trees,the results show that the intra-specific genetic distance in each barcode was lower than inter-specific genetic distance,no obvious "barcoding gap" was ob-served in every barcode,and species identification efficience of each barcode varied from 47% to 63%.Among the barcode combinations,the ITS+ycf1 has the same resolution as combining three or four barcodes,and the i-dentification efficience is up to 79%,indicating that the ITS+ycf1 barcode combination would be valuable to be selected as the standard DNA barcode for Coptis species identification.关键词
