摘要

Abstract

Objective:To study the protective effect of Bushen Huoxue Tongfu formula(BHTF)on brain tissue of rats with chronic cerebral circulatory insufficiency(CCCI). Methods:Fifty healthy male Wistar rats were randomly divided into five groups:sham operation group,model group,nimodipine group,BHTF of low dose group,BHTF of high dose group. The CCCI rat model was established using 2-VO permanent ligation. After 8 weeks of intragastric administration,the rats were sacrificed through decapitation,and the hippocampus was harvested. The protein was detected by Western blot. Results:Compared with the sham group,the expression of PGC-1α and FOXO3α in the frontal lobe of the model group and other treatment groups were decreased significantly(P<0.05). Compared with the model group,the expression of PGC-1α protein in the hippocampus of the BHTF of low dose group was significantly higher than that in the model group(P<0.05). Compared with the nimodipine group,FOXO3α,PGC-1α protein expressions were significantly increased(P<0.05). Con-clusion:Cerebral ischemia in CCCI rats can inhibit the expressions of FOXO3α and PGC-1α proteins. BHTF of low dose can improve the expressions of FOXO3α and PGC-1α proteins. Increase the blood supply of the brain,thereby protecting the brain tissue of CCCI rats.
