摘要

Abstract

Objecticve :To observe the intervening effect of Xiusanzhen Electroacupuncture(EA) on phospho-rylation of P38MAPK and TNF-α expression in hippocampus of mice with Alzheimer's disease(AD) ,in order to clarify its therapeutic mechanism based on the olfactory nerve pathway in alleviating AD .Methods :40 AD mice were randomly devided into 4 groups :AD model group ,Xiusanzhen EA with olfactory nerve transected group (ONT+EA) ,Xiusanzhen EA group ,Donepezil HCl group ,with 10 mice respectively in each group ,then took 10 nega-tive mice as normal control group .Electroacupuncture intervention of Xiusanzhen was applied to stimulate bilateral LI 20 and EX-HN3 for 10min per time ,intragastric administration of donepezil HCI in AD mice was performed at the dose of 3mg(kg · d) ,once daily intervention (except weekends) for 4 weeks in all .Western blot technique was applied to detect p-p38MAPK protein expression in hippocampus ,immunohistochemical method was used to detect the expression level of TNF-α in hippocampus .Results Compared with AD model group ,Xiusanzhen EA group Donepezil HCl group could reduce p-p38MAPK protein and TNF-α expression in hippocampus of AD mice ,there were significant differences between the two groups (P<0 .05) .There were no significant differences were found be-tween AD model and ONT+EA groups in influencing p-p38MAPK protein and TNF-αexpression in hippocampus of AD mice (P>0 .05) .There were no significant difference between Xiusanzhen EA group and donepezil HCL group (P>0 .05) .Conclusion:Xiusanzhen EA can regulate expression of TNF-αthrough p38MAPK signaling pathway ,the integrity of the olfactory pathway is the core mechanism of the intervention effect .
