摘要

Abstract

Objective To investigate the effects of miR-655-3p on proliferation,migration and invasion of gastric cancer cells. Methods The expression levels of miR-655-3p in GES-1,MKN-28,SGC-7901 and BGC-823 cell lines were detected by qRT-PCR. After transfection of agomiR-655-3p,overexpression of miR-655-3p was verified by qRT-PCR. The effect of miR-655-3p overexpression on gastric cancer cell proliferation was detected by MTT assay. The effects of miR-655-3p overexpression on gastric cancer cell migra-tion and invasion were detected by Transwell assay. Results The expression of miR-655-3p decreased in MKN-28,SGC-7901 and BGC-823 cell lines compared to that in GES-1 cell line(P < 0. 05),and the lowest in poorly differentiated BGC-823 cell line. The ex-pression level of mature miR-655-3p in BGC-823 cells increased by 35. 3 times after transfection of agomiR-655-3p(P < 0. 05). The BGC-823 cell proliferation was inhibited by miR-655-3p overexpression(P < 0. 05). The BGC-823 cell migration and invasion were de-creased by miR-655-3p overexpression. Migration cells per field decreased from 223 ± 15 to 117 ± 9(P < 0. 05),and invasion cells per field decreased from 124 ± 14 to 61 ± 6(P < 0. 05). Conclusion Overexpression of miR-655-3p can inhibit BGC-823 gastric cancer cell proliferation,migration and invasion.
