盐酸多奈哌齐片联合氯氮平分散片治疗阿尔茨海默病伴行为障碍的临床研究
Clinical trial of donepezil hydrochloride tablets combined with clozapine dispersible tablets in the treatment of elderly Alzheimer's disease with behavioral disorder

Abstract
Objective To observe the clinical efficacy and safety of donepezil hydrochloride tablets combined with clozapine dispersible tablets in the treatment of elderly Alzheimer's disease with behavioral disorder.Methods A total of 80 elderly patients with Alzheimer's behavioral disorder were randomly divided into control group and treatment group with 40 cases per group.Control group was treated with donepezil hydrochloride tablet 5 mg and increased to 10 mg after 1 month,qd,oral before bedtime.Treatment group was given clozapine dispersible tablet 25 mg for the first dose with a rate of 25-50 mg at 4-5 days within the 2-3 weeks up to 200 mg· d-1,qd,oral before bedtime,on the basis of control group.Two groups were treated for 6 months.The clinical efficacy,Alzheimer's disease behavior rating scale (BEHAVE-AD) score and adverse drug reactions were compared between two groups.Results After treatment,the total effective rates in treatment and control groups were 87.5％ (35/40 cases) and 55.0％ (22/40 cases);BEHAVE-AD scores in treatment and control groups were (4.44 ± 1.95) and (8.86 ± 2.47) points,there were statistically significant differences (all P ＜ 0.05).The adverse drug reactions in two groups were based on nausea,dry mouth,dizziness,drowsiness,weight gain and constipation.The incidences of adverse drug reactions in treatment and control groups were 40.00％ and 37.50％ without significant difference (P ＞ 0.05).Conclusion Donepezil hydrochloride tablets combined with clozapine dispersible tablets have a definitive clinical in the treatment of elderly Alzheimer's disease with behavioral disorder,without increasing the incidence of adverse drug reactions.关键词
