虎杖苷对2型糖尿病小鼠溃疡创面愈合影响的实验研究
Experimental study of polydatin on wound healing in type 2 diabetic of mice

Abstract
Objective To explore the effect of polydatin on type 2 diabetic of mice.Methods C57BL/6J male mice continued for 4 weeks after feeding with high fat diet,fasting 12 h per day,intraperitoneal injection of streptozotocin 40 mg · kg-1 weight.A continuous injection of 5 days.One week later,measurements of random blood glucose values were higher than 16.67 mmoL · L-1 for type 2 diabetic mice.Disinfection of back shaving after anesthesia,a 0.8 cm circular wound was cut on each side of the back of the mouse with a sterile surgical scissors,deep to fascia.The mice were randomly divided into experimental-H,experimental-L and control groups,15 rats in each group.The wound was locally administered on that day and served as the first day of the experiment.The experimental period for 14 days.The local concentration of experimental-H and experimental-L were 400 and 100 μg · mL-1polydatin for 5 d,take the medicine once a day,and finish it for fifth days.Discontinue medication between 6-14 days;the control group was given the same dose of 0.9% NaCl and sterile gauze.Wound healing rate and wound healing time were statistically analyzed each at 3,5,7,10 and 14 days.The 100% wound healing time were statistically analyzed.Morphological changes were observed by HE staining each at 7,14 days.Polydatin group and phosphate buffer(PBS) control group on proliferation of fibroblast chemotaxis effect were detected by MTT method and scratch test.The percentage of polarized M2 macrophage in primary macrophages was detected by flow cytometry in the polydatin group,the intedeukin(IL)-4 group,the polydatin + IL-4 group,the PBS control group.Results The wound healing rates of the experimental-H and the control were (95.93 ± 1.01) % and (80.72 ± 5.33) %,the wound healing rate of the experimental-H was significantly increased (P < 0.001).Compared with the control,the wound healing time of the experimental-H was shortened,and shortened from (16.6 ± 1.1) d to (13.2 ± 0.8) d with significantly(P <0.001).After administration 14 d,the collagen fibers in the experimental-H were arranged compactly and neatly,and the control did not heal.The 0.5 μg · mL-1 is the proliferation of fibroblasts polygonin optimum concentration in experimental group.After treating 1 d,the migration rate of polydatin group and PBS control group respectively (90.20 ± 3.90) % and (76.8 ±4.87) %,significantly contributed to the migration effect of polydatin with fibroblast cells with significantly (P < 0.01).The percentage of M2-type macrophages in the polydatin group (3.7 ± 0.36)% was significantly higher than PBS control group (1.97 ± 0.23)%,with significantly (P < 0.05).Conclusion Polydatin can promote type 2 diabetic mice ulcer wound healing.关键词
