摘要

Abstract

This experiment aimed to explore the location of nesfatin-1 protein and NUCB2 in the reproductive tissues of female dogs.Samples were obtained from puppy(1.5M),prepuberty(4M),puberty (7.5M) and adulthood (24M) female dogs,respectively.Nesfatin-1 protein distribution was determined using immunohistochemistry,andNUCB2mRNA expression in different developmental female dog reproductive axis was measured by real-time quantitative polymerase chain reaction,In result,nesfatin-1 protein was detected in the lateral hypothalamic area,supraoptic nucleus,pituitary gland,oocytes and interstitial substance of ovaries.Additionally,in pituitary and ovary,NUCB2 mRNA expression showed a significant upward trend from puppy to puberty stage(P<0.01),and swiftly decreased at adulthood (P<0.01).In the hypothalamus,there was a significant and continuing increase trend from puppy to adulthood stage(P<0.01).In conclusion,the nesfatin-1 distributed in the reproductive axis tissues widely.TheNUCB2mRNA level peaked at a puberty stage,indicating that nesfatin-1 may regulate the initial of puberty.The NUCB2 mRNA level in ovary was higher than that in the other reproductive organs,suggesting that nesfatin-i may participate in the development and function of the ovaries.
