摘要

Abstract

Objective To investigate affecting mechanism of miR-29c targeting AMPK/mTOR signaling pathway on biological behavior of gastric cancer cells. Methods The miR-29c expressions in different gastric cancer cell lines were detected by quantitative polumerase chain reaction ( qPCR) . Immunofluorescence was used to detect transfection efficiency of miR-29c-mimic transfecting SGC-7901 gastric cancer cell. MTT proliferation assay was used to detect effect of miR-29c on proliferation ability of SGC-7901 gastric cancer cell. Transwell invasive test was used to detect effect of miR-29c on inva-sion ability of SGC-7901 gastric cancer cell. Western blotting was used to detect changes of p-AMPK and PRR5 protein ex-pressions after miR-29c overexpression. Results The miR-29c expression in SGC-7901 gastric cancer cell was significantly lower than those in MGC-803, AGS and HGC-27 gastric cancer cells (P<0. 05). At 72 h after transfection, miR-29c-mimic could effectively transfect into SGC-7901 gastric cancer cell. Values of cell proliferation rate and expression levels of p-AMPK and PRR5 proteins in miR-29c-mimic group were significantly lower than those in NC group (P<0. 05), while number of cells migrated to the submucosa was significantly less than that in NC group (P<0. 05). Conclusion miR-29c may target AMPK/mTOR signaling pathway to regulate biological behavior of gastric cancer cells.
