miR-708与JAK1在大鼠脑缺血再灌注损伤中的表达及调控关系
Expression and interaction of miR-708 and JAK1 in rats with cerebral ischemia-reperfusion injury

Abstract
Objective To investigate the function and mechanism of expression changes of microRNA-708(miR-708) in rats with cerebral ischemia reperfusion injury .Methods Five male SD rats were randomly assigned to sham operation group (n=10) and tMCAO group (middle cerebral artery infarct and reperfusion model ,10 rats survived after operation ,n=10) .The miR-708 expression in brain tissues and blood and JAK 1 mRNA expression in brain tissues of each group were detected by using real time polymerase chain reaction (qRT-PCR) method .The JAK1 protein level was detected by using Western blot method .Results Compared with those in sham operation group ,miR-185 expressions were significantly lower (P<0.01) in brain tissues and blood and the expressions of JAK1 mRNA were significantly increased in brain tissues in the tMCAO group (P<0.01);Correlation a-nalysis found that the expression of miR-708 and JAK1 mRNA had negative correlation in the tMCAO rats .Conclusion MiR-708 may play a protective role in brain ischemia reperfusion injury by inhibiting the expression of the target gene JAK1 .关键词
