摘要

Abstract

Objective To evaluate the Role of mitochondrial ATP-sensitive potassium channels in attenuation of cerebral is-chemia-reperfusion injury by parecoxib in rats .Methods One hundred healthy male Sprague-Dawley rats ,weighing 280g-330g , were randomly assigned into 5 groups (n=20 each) using a random number table:sham operation group (group S);focal cerebral I/R group (group I/R);parecoxib group (group P);5-hydroxydecanoate group (group 5-HD);5-hydroxydecanoate group+ pare-coxib group (group 5-HD+ P) .Focal cerebral ischemia was produced by 2h right middle cerebral artery occlusion followed by reperfusion .Group P received parecoxib 4mg/kg intravenously 15min before ischemia and again at 12h during reperfusion ,while group S and I/R received the equal volume of normal saline instead .In group 5-HD ,5-HD 30 mg/kg was injected intravenously at 1h before ischemia .In group 5-HD+P ,5-HD 30 mg/kg was injected intravenously at 1h before ischemia and the other procedures were similar to those previously described in group P .The neurological function deficit score (NDS) was evaluated in the 5 groups in 24h after reperfusion .Then the rats were sacrificed ,and their brains were immediately removed for evaluation of the levels of su-per oxide dismutase (SOD) ,malondialdehyde (MDA) ,Bax protein and Bcl-2 protein .Results Compared with group S ,the NDS , the levels of MDA and Bax protein increased ,and the levels of SOD and Bxl-2 protein decreased in the other groups (P<0.05) . Compared with group I/R ,the NDS ,the levels of MDA and Bax protein decreased ,and the levels of SOD and Bxl-2 protein in-creased in group P (P<0.05) .Compared with group P ,the NDS ,the levels of MDA and Bax protein increased ,and the levels of SOD and Bxl-2 protein decreased in 5-HD+ P (P<0.05) .Conclusion MitoKATP channels are involved in reduction of I/R-in-duced cerebral injury by parecoxib in rats .
