心安颗粒对病毒性心肌炎小鼠心肌核因子-κB信号通路及细胞凋亡的影响
Effect of Xinan granules on the nuclear factor kappa B signal pathway and apoptosis of viral myocarditis in mice

Abstract
Objective To explore the protective effect of Xinan granule on viral myocarditis mice and its possible molecular mechanisms basing on the nuclear factor-κb(NF-κB) signal pathway and cell apoptosis.Methods Viral myocarditis (VMC) mice were induced by intraperitoneal injection of coxsackievirus B3 (CVB3).The BALB/c mices were divided into six groups according to random number table method:normal group,model group,contral group(Ribavirin,0.2 mg · g-1),stepwise dose experimental groups(Xinan granules,10,20,40 g · kg-1),20 mices of each group.The mice in model and each treatment groups were inoculated with 0.2 mL cultures containing CVB3 intraperitoneally;and those in normal control group were inoculated with 0.2 mL cultures without CVB3.After the mices were inoculated 2 h later,the normal group and model group with the same amount of saline solution for 7 d.The mices were killed on day 8 after administration.Serum cardiac troponin Ⅰ (cTnⅠ) concentration was investigated by enzyme linked immunosorbent assay.The expression level of tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6) and cluster of differentiation 68 (CD68) macrophage were detected by immunohistochemistry staining.The expression level of myocardial nuclear factor kappa B (NF-κB) p65 was determined by Western blot.Results Compared with normal group,the the serum cTnⅠ was (453.60 ± 33.85) pg · mL-1,the mean optical density of TNF-α and IL-6 were 5.25 ± 2.16 and 4.66 ± 1.28,the number of CD68 macrophages was (13.83 ± 1.94) count · mm-2 in model group with statistical difference(all P <0.01).Compared with model group,the serum cTnⅠ were (86.70 ± 14.51),(126.67 ±7.34),(104.56 ± 11.96) pg · mL-1;the mean optical density of TNF-α were 2.20 ±0.76,3.03 ± 0.59,2.78 ± 0.64;the mean optical density of IL-6 were 2.48 ± 0.52,3.39 ± 0.61,1.85 ± 0.84;the numbers of CD68 macrophages were (4.69 ± 0.53),(7.90 ± 1.95),(6.49 ± 1.44) count ·mm-2 in contral group,experimental-M,experimental-H groups with statistical difference(all P < 0.05).The expression of myocardial nuclear factor kappa B (NF-κB) p65:compared with model group (0.97 ± 0.07),these were 0.24 ± 0.02,0.51 ±0.04,0.30 ±0.07 in above three groups with statistical difference (all P < 0.01).Conclusion Xinan granule can obviously improve the myocardial inflammation and injury,and protect the myocardium of VMC mice.Its mechanisms may be associated with the inhibition of NF-κB signal pathway activation and myocardial apoptosis.关键词
