Abstract

Objective To investigate the extraction and purification process of total flavonoids from Radix Paeoniae Rubra. Methods With the yield of total flavonoids as the evaluation index, single factor and the orthogonal design were used to optimize the extraction process of total flavonoids including extraction duration, extraction temperature, solid-liquid ratio and the extraction times. D-101 resin was used to optimize the purification process of total flavonoids by measuring the adsorption ratio, eluting ratio of total flavonoids and so on. Results The optimum extract parameters were as follows, temperature of 60 ℃ , solid-liquid ratio of 1 : 12, extraction for twice and 1.5 h each time. The content of total flavonoids in Radix Paeoniae Rubra was 1.82%. The optimum purification conditions were as follows, resin-sample ratio of 1 :1, 0.5 mg/ mL sample concentration, 3 BV sample volume, 60% eluting reagent concentration, 2 BV/ h flow rate, 3 BV eluting reagent, and diameter to height ratio of 1 :9. The dry extract yield in purified substance reduced from 8.99% to 2.49% and the total flavonoids content increased from 185.76 mg/ g to 689.67 mg/ g. Conclusion The optimum extraction and purification process was economical, stable and feasible and could provide reference for the research and development of Radix Paeoniae Rubra.
