Abstract

Objective To study the protection and mechanism of proanthocyanidins on oxidative damage in SH-SY5Y cells induced by H2 O2 . Methods MTT method was used to select the useful concentration of proanthocyanidins. The model of oxidative damage on SH-SY5Y cells was established with H2 O2 ( 400μmol/ L) treatment. The cells were pretreated with 6. 25, 12. 5 and 25 μg/ mL proanthocyanidins. The changes of SH-SY5Y cell morphology were observed by inverted microscope. MTT method was used to detect the survival rate of cells, and the expression of HSPA1L protein was detected by western blot. Results Compared with the model group, the cell morphology and survival rate were significantly improved after proanthocyanidins treatment in a dosage-dependent manner. Meantime, the level of HSPA1L was significantly decreased in 25 μg/ mL proanthocyanidins group. Conclusion Proanthocyanidins can protect SH-SY5Y cells from oxidative damage induced by H2 O2 , and its mechanism may be related to modulate HSPA1L expression.
