摘要

Abstract

To further investigate the mechanism of antibiotic resistance of Salmonella enteritidis (S.enteritidis),the specific primers were designed,based on sdf Ⅰ of S.enteritidis from the GenBank.A PCR method for S.enteritidis detection was developed after optimizing reaction conditions,and used for detection of clinical S.enteritidis isolated from chickens.The isolates were evaluated for antimicrobial sensitivity by K-B disc diffusion method against 22 antimicrobial drugs and the resistance genes of the isolates were detected by PCR.Electrophoresis showed that 203 bp fragment was detected,and the PCR assay could detect 100 pg/μL of DNA sensitivity,and 4 × 103 cfu/mL of the bacteria sensitivity,respectively.Compared with the traditional serological assay,the coincidence rate of the PCR for the isolates were 100%.The results of drug sensitive test showed that 56 isolates were resistance to the 22 antimicrobial agents in some degree,and penicillin (98.21%) and erythromycin (96.43%) were the highest.Test of resistance genes exhibited that the detectable rate of gene tetA,aph (3')-Ⅱ a,blaCMY-2,cat1 and sul1 was higher(more than 50%).In conclusion,the developed PCR assay was more specific,sensitive and rapid for the detection of S.enteritidis,and was able to be used for the rapid identification of S.enteritidis and epidemiological investigation.The resistance analysis results showed that the problem of resistance was serious,and resistance genes were widely existed in the resistant strains.
