Abstract

Objective: To explore the mechanism of salidroside in improving insulin resistance in the skeletal muscle. Methods:The type Ⅱ diabetic rat model was established with a small dose of STZ plus high-fat and high calorie feeding. The modeled rats were randomly divided into diabetic model (DM) group, salidroside low-dose group, salidroside middle-dose group, salidroside high-dose group and metformin group. Another 10 rats were set as normal control group (NC). Blood glucose, blood lipid and insulin levels were measured after 12 weeks. The rats were sacrificed and the hind legs were removed. Western blotting was used to detect the expression levels of PI3K and GLUT4 in tissues. Results: Compared with NC group, the expression of PI3K and GLUT4 in skeletal muscle cells of rats in DM group was significantly reduced. Compared with DM group, the expression of PI3K and GLUT4 in skeletal muscle cells was significantly enhanced in drug treatment group. Conclusion: Salidroside may improve insulin resistance in type Ⅱ diabetic rats by increasing the expression of PI3K and GLUT4 in rat skeletal muscle cells.
