Abstract

Microbial communities inhabiting in soils are one of the crucial players in maintaining health and balance of the soil ecosystem.The impact of different land use and fertilization on soil microbial community structure is significant.This study aimed to evaluate the microbial community structure and diversity in upland and paddy soil which with fertilization (US/PS-NPK) or without fertilization (US/PS-CK).These soils have been managed for 20 years.Using the 16S rRNA-based pyrosequencing,we compared and analyzed the microbial community structure and diversity,and did correlation analysis combining with environmental factors.The results showed that the order of diversity of microbial communities was as follows:US-NPK < US-CK < PS-CK < PS-NPK,which indicating the shift of upland soil to paddy soil increased the microbial diversity,and the influence of land use change on microbial diversity is significantly higher than that of long term fertilization.The dominant flora of the four kinds of soils is consistent with normal soil,which are Chloroflexi (22.0% ~30.3%),Proteobactria (18.2% ~32.7%),Acidobnctria (19.7% ~ 24.3%),and Actinobnctria (6.0% ~ 13.6%).However,the metabolic function of the microbial communities in the four kinds of soil showed difference.The study further revealed the responding mechanism of soil microbes under ecological environment change,,and provides the scientific basis for the improvement of fertilization and farming system
