视黄醇代谢相关基因LRAT和RDH12在宫颈鳞癌细胞系中的抗肿瘤作用
The effect of retinol metabolism related genes LRAT/RDH12 in the progress of squa-mous cervical cancer cell line

Abstract
Objective:To investigate the effects of metabolism-related genes LRAT and RDH12 in cervical squamous cell carcinoma cell lines SiHa and CaSKi on cell proliferation,ap-optosis,cell invasion and migration. Methods:LRAT/RDH12 over expression plasmids were carried to SiHa and CaSKi cell lines,respectively. CCK-8 method was used to detect cell prolif-eration,further through the Annexin V-FITC ( green fluorescence) and propyl iodide organism ( PI) ( red fluorescence) dyeing transfection cells to detect the cell cycle and cell apoptosis. Tr-answell chamber was used to detect the migration and invation of cervical squamous carcinoma. Results:We successfully constructed LRAT and RDH12 expression vectors and transfected cells. In the G0/G1 phase SiHa and CaSki cell lines accounted for a higher proportion than the control group,and in the S phase the proportion was lower. Compared with the control group,the percentage of apoptotic cells in SiHa cell lines increased by about 11% and 10%,respectively, and in CaSki cell lines,both groups were increased by about 7%. In the SiHa and CaSki cells, group transfected with LRAT and RDH12 , the number of the migration and invasion was less than the blank plasmid transfected group. Conclusions:By constructing LRAT and RDH12 ex-pression vector and successfully transfection cells,it was suggested that these two genes had in-hibitory effects on proliferation,apoptosis,and tumor invasion and migration in cervical cancer cell lines. It is suggested that this gene may be the tumor suppressor gene of cervical cancer.关键词
