TACSTD2在卵巢高级别浆液性癌中的表达及意义
Correlation betweene levated TACSTD2 expression in high-grade ovarian serous carcinoma and tubal origin carcinogenesis

Abstract
Objective To explore the abnormal expression of tumor associated calcium signal transducer 2 (TACSTD2) in high-grade ovarian serous carcinoma and to reveal its correlation with tubal origin carcinogenesis .Methods Thirty-six tissue specimens were respectively collected from high-grade ovarian serous carcinoma , fallopian tube epithelium , and ovarian surface epithelium . Expression of Müllerianorig in marker paired box gene 8 (PAX8) was detected by immunohistochemistry ,and histologic phenotype similarity was compared in three groups .Gene expression profile was compared in three groups using ILLUMINA Beadchip ,and target gene highly correlated with high-grade ovarian serous carcinoma was screened .Expression difference of target gene in various tissues was detected by real time polymerase chain reaction (RT-PCR) and Western blot .Results Positive expression rate of PAX8 was 100 .00% (36/36) in fallopian tube epithelium ,83 .33% (30/36) in high-grade ovarian serous carcinoma and 16 .67% (6/36) in ovarian epithelium .Difference in former two was not significant (χ2 = 2 .180 ,P > 0 .05) ,but difference in latter two had statistical significance (χ2 = 10 .670 , P < 0 .05) ,which indicated histologic phenotype similarity of former two .Beadchip showed that gene expression profile in high-grade ovarian serous carcinoma and fallopian tube epithelium was more similar compared with ovarian epithelium .Screened target gene TACSTD2 was proved to have increased expression in gene and protein level in high-grade ovarian serous carcinoma than in fallopian tube epithelium (t = - 50 .411 ,P < 0 .05) .Conclusion The high expression of TACSTD2 in high-grade ovarian serous carcinoma might associate with tubal origin carcinogenesis .关键词
