Abstract
Objective To investigate the relationship between polymorphism of PD-1 .1 (rs36084323) and PD-1 .5 (rs2227981 ) in programmed apoptosis protein 1 (PD-1 ) and susceptibility and prognosis of epithelial ovarian carcinoma (EOC ) .Methods Altogether 116 patients with EOC (case group) receiving treatment in Rizhao Hospital of Traditional Chinese Medicine from April 2009 to May 2012 were collected ,and 110 healthy subjects receiving physical examination at the same time were selected as controls (control group ) .Blood samples in two groups were collected .RT-PCR was used to analyze the relationship of polymorphism of PD-1 .1 and PD-1 .5 in PD-1 with EOC and prognosis .Results Genotype distribution and allele frequency of PD-1 .1 in case group were significantly different from those in control group (χ2 value was 7 .785 and 7 .970 ,respectively ; P value was 0 .020 and 0 .005 ,respectively ) .Frequency of PD-1 .5 allele in case group was significantly different from that in control group (χ2 = 4 .939 ,P = 0 .045) .PD-1 .1 genotyping was correlated with clinical stage and tumor differentiation of EOC patients (χ2 value was 7 .744 and 10 .229 ,respectively ;P value was 0 .021 and 0 .006 ,respectively) .Recurrence rates of 3-year and 5-year in AA type , AG type and GG type of PD-1 .1 were 70 .27% ,68 .42% ,36 .36% and 86 .49% ,80 .70% and 63 .64% ,respectively .Differences in recurrence rates of 3-year and 5-year in three genotypes were statistically significant (χ2 value was 11 .634 and 8 .218 ,respectively ;P value was 0 .003 and 0 .011 ,respectively) .Five-year survival rates in PD-1 .1 gene AA type ,AG type and GG type were 18 .92% , 43 .86% and 54 .55% ,respectively ,and Kaplan-Meier analysis showed statistically significant difference (χ2 = 9 .111 , P = 0 .011) . Multivariate regression analysis by Cox showed that PD-1 .1 genotype was risk factor for 5-year recurrence and mortality in EOC patients (HR = 2 .709 ,95% CI :1 .127-4 .892 ,P = 0 .004) .Conclusion G gene carriers in PD-1 .1 polymorphic locus may significantly reduce risk of EOC .C and T genes of PD-1 .5 polymorphic locus may not be associated with the pathogenesis of EOC .PD-1 .1 polymorphic locus may be associated with clinical outcomes in patients with EOC .关键词
