农杆菌介导的玉米幼胚遗传转化体系的建立

Abstract
With maize immature embryos as receptor materials,the plant expression vector pCAM-BIA1300-35S-GUS-Nos was introduced into maize immature embryos by Agrobacterium-mediated method.With GUS transient expression efficiency as indicator,the factors,such as,genotype,immature embryo size,infection liquid concentration and infection time,were studied by single factor test and the L9(34) orthogonal experiment to explore their effects on genetic transformation rate.The results show that:The effect rank of these factors was genotype>embryo size>bacterial concentration >infection time;The optimal infection rate was the genotype H99,the size of immature embryos was 1.1-2.0 mm,OD600 =0.6,the time of infection was 30 min,and the transient expression rate of GUS gene was (34.44± 1.11) %.关键词
