摘要

Abstract

Objective The objective of this study was to develop a borate bone cement (BBC) that could serve as a carrier for delivering Gentamycin Sulfate (GS) and evaluate the capacity of antibiotic in vitro.Methods A local drug delivery system,using novel BBC as carrier for GS,was prepared.The effect ofdrug loading on the injectability,initial setting time of the injectable BBC was tested.The bioactivity,biodegradation and loading/release capacity of the BBC was evaluated by immersing the pre-set BBC samples in phosphate buffered saline,PBS.The in vitro antibiotic property was test by the inhibition zone of the E.coli and S.aureus cultured on the LB agar plates.Results The BBC loaded with GS can be almost completed injected by syringe,and had a proper initial setting time of about 6 min.When immersed inPBS,sustained release ofGS occurred over 26days,while the borate bioactive glass in BBC converted continuously to hydroxyapatite (HA).After loading with GS,the BBC can inhibit the growth of E.coli and S.aureus effectively.Conclusion The results suggest that,GS loaded BBC,had an excellent injectability,proper in-situ self-curing time,long and sustained drug release and the ability to inhibit bacterium growth.
