Effects of high progesterone in late follicular phase on pinopode and the expression of integrin, LIF and IL-1 in endometrium of patients with normal ovarian responses

Abstract
Objective To explore the changes of pinopode and the expression of integrin,LIF and IL-1 in endometrium under high levels of progesterone in late follicular phase in patients with normal ovarian responses.Methods This was a prospective analysis conducted in the Reproductive Medical Center of the Second Affiliated Hospital of Zhengzhou University.Totally 29 patients with normal ovarian responses were collected,who received long agonist program treatments in IVF-ET cycles and canceled embryo transfer from April 2014 to December 2015.Patients were divided into two groups according to their serum progesterone concentration on the day of hCG administration:≥7.13nmol/L (high P group,n=18) and ＜7.13nmol/L (normal P group,n=1 1).Endometrial biopsies were performed 5 days after oocyte retrieval.The changes of pinopodes were observed by scanning electron microscope.The expression of integrin,LIF and IL-1 was detected by Western blot method.Results The pinopodes were wrinkled in the high P group,but in the normal P group they were in a state of development and maturation.The expression of integrin (0.227±0.060 vs.0.512±0.031),LIF (0.144±0.018 vs.0.474±0.041) and IL-1 (0.220±0.009 vs.0.460±0.035)in the high P group were lower than the normal group (P＜ 0.05).Conclusion High P level on the day of hCG administration may affect the expression of pinopodes and integrin,LIF and IL-1 in the peri-implantation period of these patients with normal ovarian responses.The elevated progesterone may lead to the early maturation of the endometrium during implantation phase,leading to a failure of the embryo-endometrium dialogue.关键词
