Abstract
Objective To investigate the correlation between SHBG gene (TAAAA)n polymorphism in peripheral blood and the incidence of gestational diabetes mellitus (GDM).Methods Totally 211 GDM patients diagnosed in outpatient clinic from Jan 2011 to Feb 2016 in Shengjing Hospital of China Medical University were enrolled in our study as GDM group;205 healthy pregnant women giving birth to girls at the same period were selected as the control group.Gene fragment analysis and sequencing method were used to detect the SHBG (TAAAA)n gene polymorphism and repetition frequency.Enzyme-linked immunosorbent assay (ELISA) method was used to detect the serum SHBG level in peripheral blood and umbilical blood.SPSS20.0 software was used to make data analysis.Results Five kinds of allelic genes [(TAAAA)n 6,7,8,9,10 repetition were detected in peripheral blood of all pregnant women in both groups,14 combined genotypes in total.The peripheral blood SHBG (TAAAA)n gene detection showed there was statistical difference in the distribution of 5 allelic genes (P=0.004) between GDM group and control group,the proportion of 8/8 and 9/9 allelic genes being higher in GDM group (P=0.027,P=0.041) than in control group.The serum SHBG level of long allele genotype was lower than short allele genotype in maternal peripheral blood in GDM group (P＜0.001).Conclusion In the SHBG(TAAAA)n gene polymorphism the women with 8/9 and 9/9 allelic genes are more susceptible to GDM,who are the high-risk population.Gene detection of SHBG (TAAAA)n in peripheral blood can be a clinical reference for GDM incidence in pregnant women.关键词
