摘要

Abstract

Objective To observe the effect of Platycodon grandiflorum saponins(PGS) on the expression of surfatcant - associated protein A ( SP - A ) in rats with lung injury induced by PM2. 5, and to investigate its repair effect on lung injury induced by PM2. 5. Methods Totally 50 Wistar rats (male and female in half, weighing 180 - 220 g) were randomly divided into 5 groups: blank control group, model control group, PGS high, middle and low dose group, 10 rats in each group. The models were established by tracheal instillation of PM2. 5. After administration of 14 d, the middle lobe of left lung tissues and right lung tissues were taken and stained with conventional HE, the pathological changes of lung tissue were observed under microscope. The expression of SP - A in the lung tissues was detected by Western blot(WB), the expression of mRNA in the lung tissues was detected by real - time quantitative PCR (RT - PCR) . BALF and serum of the rats were collected, and the content of SP - A in BALF and serum were determined by ELISA. Results PGS can significantly inhibit inflammation of lung tissue that damaged by PM2. 5, increase the expression of SPA in lung tissue and the content of SPA in BALF, but decrease the content of SPA in serum of rats. Conclusion PGS can repair the type Ⅱalveolar epithelial cells(AEC - Ⅱ) and capillary in lung tissue that damaged by PM2. 5.
