摘要

Abstract

Objective To observe the impact of umbilical cord-derived mesenchymal stem cells (UCMSCs) treatment on the expression of Th1/Th2 cytokines in peripheral and splenic lymphocytes of tree shews with radiation injury.Methods Adult male tree shrews were randomly divided into a model group and a treatment group (n=15,respectively);both groups received one-time ionizing radiation of 4.5 Gy to the whole body;and then,the treatment group received an infusion of allogeneic UCMSCs (3×107 cells/kg) via the femoral vein and the model group was treated with normal saline of the same volume,and both groups were treated for four consecutive times,once per week.One week after the last MSC treatment,the statistical treatment was done on the survival rate of both groups,and then the real-time quantitative polymerase chain reaction (PCR) technique was performed on the peripheral and splenic lymphocytes in both groups to obtain the mRNA expression levels of IL-2,IL-4,IL-6 and TNF-α.Results One week after the last treatment,the survival rate of the treatment group was 86.677% and that of the model group was 53.33%,presenting a significant difference (P < 0.05);the expression levels of TNF-o and IL-2 in the peripheral and splenic cells of the treatment group were lower than those in the model group (P < 0.05),while the expression levels of IL-4 and IL-6 were higher than those in the model group (P <0.05).Conclusion The UCMSCs treatment can improve the survival rate of tree shrews with radiation injury to the whole body and induce a switch of the Th1/Th2 cytokine expression in lymphocytes to the dominant Th2 cytokine expression.
