BMSC治疗ApoE-/-动脉粥样硬化小鼠模型的初步研究
A preliminary study on effect of BMSC on atherosclerosis in ApoE-/-mouse model

Abstract
Objective To study the therapeutic effect of bone marrow-derived mesenchymal stem cells (BMSCs) on the atherosclerosis (AS) in ApoE-/-mice.Methods ApoE-/-mice were randomly divided into a blank control group,a negative control group,an experimental group and a normal control group.From the ninth week,the experimental group and the negative control group were injected with 200 μl (1×105 cells) BMSC suspension via caudal vein and normal saline of same volume respectively,while the normal control group and blank control group were not treated.In the 12th week and after four weeks since the BMSC injection,the levels of total cholesterol (TC),triglyceride (TG),high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C) in the blood of all groups were measured by an automatic biochemical analyzer,and the oil red O staining was conducted to observe the formation of plaques in the aortic arch.Results As compared with the normal control group,the levels of TC,TG and LDL-C in the blank control group increased significantly(P < 0.05);the oil red O staining showed the presence of plaques in the aortic arch of the blank control group.After BMSC treatment,the levels of TC,TG and LDL-C in the experimental group were much lower than those in the negative control group (P < 0.05);the oil red O staining suggested less and smaller plaques in the aortic arch of experimental group.Conclusion ApoE-/-mice fed a high fat diet till the 12th week showed clinical symptoms of typical atherosclerosis (AS),and the allogeneic BMSCs play a role in reducing blood lipids and inhibiting the formation of plaques in the aortic arch.关键词
