摘要

Abstract

Objective To observe the effect of umbilical cord-derived mesenchymal stem cells (UCMSCs) on CD4+ CD25+ Treg cells in vitro.Mthods Human UCMSCs and lymphocytes were isolated and co-cultured in different proportions in contact and noncontact modes for 72 h;additionally,other lymphocytes were isolated and cultivated to be a control group.Lymphocytes of all groups were collected and treated with the flow cytometry to measure the percentage of CD4+ CD25+Treg cells;and then,the supernate of all groups was obtained and treated with the enzyme-linked immunosorbent assay (ELISA) method to measure the level of IL-10.Results When lymphocytes were co-cultured in contact and non-contact modes with UCMSCs in the proportion of 10:1,Treg cells accounted for the highest percentage among all cells;the levels of CD4+ CD25+Treg cells and IL-10 in all co-culture groups were significantly higher than those in the control group (P < 0.05).Conclusion UCMSCs could induce lymphocytes to differentiate into Treg cells;the mechanism of such differentiation may be related to cytokines released by UCMSCs.
