Abstract
Objective To explore the expressions of circulating miR-26a and its target gene PTEN in systemic lupus erythematosus (SLE) and their possible roles in the occurrence and development of SLE.Methods The expressions of circulating miR-26a and PTEN in peripheral blood mononuclear cells (PBMCs)were detected by qRT-PCR after blood samples were collected from 36 cases of SLE and 10 healthy people.The correlations between miR-26a,PTEN and clinical data of SLE were statistically analyzed.Dual luciferase reporter system was used to test whether PTEN was the target gene of miR-26a.Results The expression of miR-26a mRNA in the serum of the SLE patients was higher than that in the healthy people (P< 0.05),and the expression of PTEN mRNA in the PBMCs of the SLE patients was lower than that in the PBMCs of the healthy people (P < 0.05).The expression levels of miR-26a and PTEN in the patients with SLE were correlated with C3,anti-dsDNA,ESR and SLEDAI.After miR-26a was transfected to BaF3 cells,the expression of PTEN was down-regulated,and the expression of PI3K was up-regulated,and the luciferase activity of the transfected group was decreased (P < 0.05).Conclusions The over-expression of circulating miR-26a may participate in the occurrence and development of SLE through down-regulation of its target gene PTEN and up-regulation of PI3K/AKT signaling pathway.关键词
