摘要

Abstract

Objective:To prepare the biomimetic structure of Tanshinone Ⅱ A albumin linked lipid core by simulating the supramolecular structure of native lipoproteins.Methods:Tanshinone Ⅱ A loaded nanostructured lipid canriers was prepared by emulsification-evaporation method,and then the bovine serum albumin was incabated by physical method,bionic carrier structure which approach to the natural lipoprotein morphology.The particle size and encapsulation efficiency were used as indexes to choose the best prescription of Tanshinone Ⅱ A nanostructured liposomes.By fluorescence quenching method,the temperature,time and the amount of the bovine serum albumin of the incubation condition were observed.Results:The optimum process condition was obtained,the size date of the best Tanshinone Ⅱ A loaded nanostructured lipid carriers was (42.1 ± 2.1) nm,the encapsulation efficiency of the formulation was (100.60 ± 1.93)%,the Zeta potential of the best formulation was(-28.52 ± 0.34)mV;the particle size of the best prescription incubated with bovine serum albumin was(185.3 ± 1.2)nm,the Zeta potential of the best prescription incubated with the bovine serum albumin was (-18.26 ± 0.70)imV.The images received from the Transmission electron microscope showed that the bovine serum albumin was incubated to the tanshinone Ⅱ A which loaded nanostructured lipid carriers successfully.Conclusion:The prepared Tanshinone Ⅱ A loaded nanostructured lipid carriers has uniformly small size distribution and high encapsulation efficiency.And after being incubated with the bovine serum albumin,the carriers shows the basic form of superamolecular structure which is similar to nature lipoprotein.
