Abstract

Objective:To investigate the effects of exogenous AgNO3 on the growth and gentiopicroside accumulation of Gentiana macrophylla seedlings.Methods:Gentiana macrophylla seedlings were used to reserch,the concentrations of gentiopicroside in Gentiana macrophylla seedlings treated with 1 mmol/L AgNO3 were measured by HPLC method.The growth of the Gentiana macrophylla seedlings were observed after treated with AgNO3.The expression of key enzyme geneson and the gentiopicroside biosynthesis pathway were detected by Real time fluorescent quantitative PCR(RT-qPCR).Results:Compared with control group,the content of gentiopicroside in Gentiana macrophylla seedlings increased to 1.91 times,and the fresh weight,dry weight and root length were increased by an average of 29%,32% and 43% when they were treated with 1 mmol/L AgNO3 for 9 d.RT-qPCR results showed that the expression of some key enzyme geneson gentiopicroside biosynthesis pathway were significantly enhanced after treated with exogenous AgNO3.Conclusion:Exogenous AgNO3 can significantly promote the accumulation of gentiopicroside in Gentiana macrophylla seedlings,increase the biomass(dry weight and fresh weight)and root elongation of Gentiana macrophylla seedling.
