Abstract

Objective:To establish the UPLC fingerprint of Scutellaria indica,and infer the structure of the chemical composition in Scutellaria indica using LC-MS technology.Methods:UPLC method was taken on Agilent Poroshell 120 SB-C18 column(100 mm× 2.1 mm,2.7 μm),and the mixture of 0.1% formic acid(A)-acetonitrile(B) were used as the mobile phase in a gradient elution model;the wavelength was 275 nm,the flow rate was 0.4 mL/min,the temperature was 35 ℃ and the sample size was 2 μL.In the positive and negative ion mode,the Triple TOF 5600 detector and the ESI source were used to determine the parameters of the mass spectra:the mass scan range was m/z 50 ~1 250 Da;the source temperature was 500 ℃;the atomization gas was 55 psi;the auxiliary gas was 55 psi;the air curtain gas was 35 psi;the ion scanning voltage was 5 500 V (positive ion mode),-4 500 V (negative ion mode).Results:32 common peaks were confirmed from twelve batches of Scutellaria indica UPLC fingerprint.Combining with the reference substance,literature and according to the corresponding fragments of cracking rule infference,27 flavonoids were determined.Conclusion:UPLC fingerprint of Scutellaria indica is established and the chemical components are identified by high resolution mass spectrometry,from the point of view of qualitative controls the quality of Scutellaria indica.
