miR-26a与EZH2蛋白的关系及其对肺癌细胞的增殖和凋亡行为的影响
Relationship between miR-26a and EZH2 Protein and Their Effects on Proliferation and Apoptosis of Lung Cancer Cells

Abstract
Objective To investigate effect of miR-26a on proliferation and apoptosis of lung cancer cells by in-hibiting EZH2. Methods The miR-26a expressions in lung cancer tissues and adjacent normal tissues were detected by quantitative polymerase chain reaction (qPCR). Relationship between miR-26a and EZH2 was detected by double lucif-erase assay. Effect of miR-26a on proliferation of lung cancer cells was detected by MTT assay. Effect of miR-26a on ap-optosis of lung cancer cells was detected by apoptosis examination. Results The miR-26a expression in lung cancer tis-sues was significantly lower than that in adjacent normal tissues (P<0. 05). The miR-26a expression in miR-26a mimics group was significantly higher than that in control group, while miR-26a in miR-26a inhibitor group was significantly lower than that in control group (P<0. 05). At 72 h and 96 h after transfection, proliferation activity in miR-26a inhibitor group was better than that in control group, while at 96 h after transfection, proliferation activity in miR-26a mimics group was worse than that in control group (P<0. 05). Values of apoptosis rate in miR-26a mimics group were significantly higher than those in control group, while the values in miR-26a inhibitor group were significantly lower than those in con-trol group (P<0. 05). EZH2 protein expression in miR-26mimics group was significantly lower, while the level in miR-26 inhibitor group was significantly higher than those in control group (P<0. 05). Conclusion The miR-26a can affect proliferation and apoptosis of A549 lung cancer cells by regulating expression of EZH2 protein.关键词
