摘要

Abstract

RPA(recombinase polymerase amplification) assay for detection of type A influenza virus was developed based on M gene nucleotide sequence of type A influenza virus.We verified the designed primers and probes through the specific selection of common pathogens of animal diseases.Then multiple pairs of primers were designed to select optimal reaction system and reaction conditions,and to compare with real-time RT-PCR method in sensitivity.The results showed that the RT-RPA method had strong specificity.It was rapidly tested about 20 minutes for detecting influenza A virus.The detection limit of RT-RPA for virus genome DNA was about 0.96 × 105 μg/mL,slightly lower than the detection limit of real-time RT-PCR.Thus the RT-RPA method detection of type A influenza virus is specific,sensitive,simple,rapid,and more suitable for use in field conditions and primary level laboratories.
