快速检测奶牛乳房炎四种病原菌的多重PCR方法的建立及应用
Development and applications of a multiplex-PCR for quick detection of four pathogens resulting in bovine mastitis
摘要
Abstract
Bovine mastitis is one of the major diseases that damage the economic benefits and development of dairy industry in the world,and the detection method is complicated.To accomplish rapid and accurate diagnosis of mastitis,a sensitive and special detection method is necessarily established.According to the 16S to 23S rRNA space regions of Escherichia coil,Staphylococcus aureus and Strepotococcus agalactiae and 18S rRNA sequence of Candida albicans published on GenBank,4 pairs of specific primers were designed in this study to establish a multiplex polymerase chain reaction(PCR) detection method for the four pathogens.The results showed that the positive bands of four bacteria were amplified,respectively,and the multiplex PCR method had good specificity to detect E.coil,S aureus,S.agalactiae and C.albicans,and the sensitivity rates were 1× 102 CFU/mL,1× 104 CFU/mL,1× l05 CFU/mLand 1× l02CFU/mL,respectively.关键词
