Abstract

Transcriptome analysis on Mycoplasma ovipneumoniaeNM-151 was applied for optimizing the culture medium ofMycoplasma.Samples of NM-151 strain were collected in the logarithmic phase,stationary phase and decline phase for the analysis of expression level of genes and KEG Gpathway enrichment analysis after transcriptome.Based on the above results,four schemes of medium were optimized and further proved via culture test:theNM-151 strain was incubated in four media(1:20) for 240 h,and bacterial suspensions were collected every 12 h to make the growth curve of Mo using color change unit (CCU),colonyforming unit(CFU),pH values andD~0values.The results showed that expression level of purine nucleoside phosphorylase(PNPase) and pyrimidine nucleoside phosphorylase(PyNPase) showed a downward trend over time,and the former showed significant higher expression level(P<0.01).Basal medium with the addition of serine exhibited no obvious effects on the growth of MoNM-151 (P>0.05),and basal medium with the addition of 20mmol/L HEPES extended the stationary phase and improved the microbial content (P>0.05).Moreover,basalmedium with the two additions mentioned above extended the stationary phase to 240 h and improved the microbial content from 108 to 109CFU/mL(P<0.01).Serin-HEPES modified Hayflick's could prolong the stationary phase of Mo NM-151 and improved the microbial contents,which provides references for further studies on Mo.
